Mesh biomaterials have become the standard in the treatment of hernias, regardless the location. In addition to the obvious advantages of the methods based on implantable biomaterials, one should be aware of the possible complications, such as their migration to the abdominal organs. Material and methods. The study group comprised patients operated at the Department of General, Gastroenterological Oncology, and Plastic Surgery during the period between 2008 and 2011, due to hernia surgery with mesh implantation. We also analysed the number of patients operated, due to complications of mesh migration during the same period. Results. 368 patients were subject to mesh implantation, due to hernias during the period between 2008 and 2011. Three patients underwent surgery because of symptomatic migration of the mesh (ileus, fistula). Conclusions. The frequency of mesh migration is difficult to determine because of the different criteria of migration, observation period, and other factors. In patients after mesh implantation the potential migration of the biomaterial should be considered in case of unclear or acute abdominal symptoms.

The paper presents investigation results of the migration of a chemical compound contained in fly ash deposited on a dry furnace waste landfill site exposed to weather conditions. Climate conditions are able to significantly affect chemical component distribution in a block of deposited, moving chemical compounds to different depths. The main aim of the investigations was to determine the chemical component distribution of deposited fly ash in the landfill. Identification of chemical components based on XRF analysis indicated the existence of differences in both tested storage layer and the fraction of fly ash.

Ecto-5'-nucleotidase (eN, CD73) mediates extracellular adenosine production from 5'-AMP. Non-enzymatic functions of eN have also been reported. The aim of the study was to investigate the role of ecto-5'-nucleotidase in aggressive melanoma behaviour. Analysis of the involvement of eN in adhesion, migration and invasion revealed eN functions unknown to date. We found that following eN blockade by concanavalin A, the strength of adhesion to different ECM proteins was not altered, but at the same time the invasion ability of the cells was decreased. On the other hand, knocking down eN in melanoma cells did not influence cell invasion but abolished their migration on tenascin C (TnC). Ecto-5'-nucleotidase seems to fulfil a more distinct role as a receptor than as an enzyme in the cell interaction and mobility on TnC. Ecto-5'-nucleotidase activates also focal adhesion kinase and enhances the formation of complexes upon cell adhesion to TnC. All these observations prove that an eN-TnC complex is involved in cell migration and invasion and thus in the regulation of melanoma progression.

Earlier reports suggest that the endocannabinoids may play a role of endogenous tumor growth modulators. In this study, we investigated whether inhibition of the enzymes involved in the synthesis and degradation of endocannabinoids may reduce colorectal cancer cell invasion and migration. The human colon adenocarcinoma Colo-205 cells were incubated with PF-3845, JZL-184 and RHC-80267 (fatty acid amide hydrolase (FAAH), mono- (MAGL) and diacylglycerol lipase (DAGL) inhibitors, respectively) for 48 h. The MTT colorimetric assay was performed to quantify cell viability. Next, Colo-205 cells were incubated with PF-3845 alone or with PF-3845 together with selected antagonists: AM 251, AM 630, SB 366791, RN 1734 and G-15 (CB1, CB2, TRPV1, TRPV4 and GPR30 antagonists, respectively). Western blot assay was applied to identify the changes in CB1 and CB2 receptor expression. Migration and invasion assays were employed to characterize the effect of PF-3845 on colorectal cancer cell invasion. We found that of all the inhibitors used, the FAAH inhibitor PF-3845 reduced the Colo-205 cell line viability the most effectively (IC50=52.55 μM). We also showed that the effect of decreased cell viability was enhanced when Colo-205 cells were incubated with PF-3845 and RN-1734, a TRPV4 antagonist (IC50=30.54 μM). Western blot assay revealed significantly decreased CB1 receptor expression levels, while CB2 expression was increased in response to PF-3845 when compared to control. Furthermore, PF-3845 inhibited migration and invasion of Colo-205 cell line. These results suggest that pharmacological inhibition of FAAH and consequent enhancement of the endocannabinoid levels may reduce the colorectal cancer growth and progression.

The metastatic transformation of melanocytes is associated with altered expression of adhesion molecules, including αvβ3 and α3β1 integrins. Integrin αvβ3 is a primary vitronectin (VN) receptor, while both integrin types take part in adhesion to VN when they are in complex with uPAR. Although their role in melanoma cell interaction with VN is of great interest, the influence of N-oligosaccharides attached to these glycoproteins is still unappreciated. The present study assesses the role of αvβ3 and α3β1 integrins and the influence of their glycosylation status on WM9 and WM239 metastatic melanoma cell interactions with VN. Cell adhesion to and migration on VN were selected as the studied cell behaviour parameters. Functionblocking antibodies and swainsonine (SW) treatment were used in these tests. Both cell lines interacted with VN in an integrin-mediated but cell-line-specific manner. In WM9 cells, migration was not completely inhibited by antibodies against α3β1 or αvβ3 integrins, suggesting the participation of other VN receptors. In both cell lines in coprecipitation test the formation of an integrins/uPAR complex was shown. In the presence of SW formation of the complex did not occur, suggesting the participation of glycosylation in this proccess. Additionally, the adhesion properties of WM9 cells were changed after SW treatment. Our results suggest that in these two metastatic cell lines integrin-linked N-oligosaccharides influence the VN adhesion receptor activity and function.

Стаття присвячена дослідженню особливостей еміграції високоосвітченої молоді з України, зокрема, звязку між різницею в рівнях економічної свободи, що включає вищий рівень оплаті праці та добробуту. За допомогою опитування отримано інформацію щодо впливу рівня економічної свободи на рішення щодо еміграції та вибору країни-реципієнта. У результаті аналізу результатів опитування через призму важливості економічної свободи та її значення для респондентів, автори дійшли до висновку, що економічна свобода насправді виступає важливим фактором для прийняття рішення про еміграцію.

This paper is based on the idea that there is a special pattern in migration of the well-educated youth from Ukraine connected with the difference in levels of economic freedom that include higher salary and welfare. The survey was conducted in order to obtain data regarding influence of the level of economic freedom on the decision to leave and choice of the target country. The analysis of the survey results was carried out in terms of economic freedom and its importance to the respondents, that has brought authors to the conclusion that economic freedom is indeed important factor for educated youth when the decision to leave is made.

Regiony peryferyjne są powszechnie uważane za obszary wymagające wsparcia rozwojowego. Tradycyjne metody wspierania rozwoju nie przynoszą oczekiwanych rezultatów, konieczne jest więc poszukiwanie metod alternatywnych. Skutecznym narzędziem może być prowadzenie polityki migracyjnej zróżnicowanej regionalnie i ukierunkowanej na wzrost najważniejszego z kapitałów rozwojowych – kapitału ludzkiego. Kluczowym elementem tej polityki powinien być system zachęt administracyjnych dla imigrantów nakłaniający ich do pozostania w regionach peryferyjnych lub do przeprowadzenia się tam z innych obszarów kraju imigracji. Przedmiotem szczegółowej analizy jest polityka migracyjna Polski.

The peripheral regions are areas in need of development support. Traditional methods of support do not bring the expected results, so it is necessary to look for alternatives. This research shows that a regionally differentiated migration policy oriented to the increase in human capital may be a good choice, with a system of administrative incentives for immigrants designed to encourage immigrants to stay in the peripheral regions or to move there from other areas of the country of immigration. The subject of detailed analysis is the migration policy of Poland.

Ruch i migracja są jedną z głównych funkcji życiowych komórek. W odpowiedzi na różne bodźce, dynamiczny cytoszkielet aktynowy generuje siłę umożliwiającą komórce przemieszczanie się w trójwymiarowej sieci zewnątrzkomórkowej macierzy czy po płaskim podłożu. Wydłużanie filamentów aktynowych na ich kolczastych końcach wypycha błonę komórkową w kierunku migracji, formując strefę frontalną zwaną lamellipodium. Skurcz włókien naprężeniowych umożliwia oderwanie tylnej części komórki i przesunięcie jej do przodu. W odpowiedzi na bodźce ze środowiska, receptory komórki inicjują wiele szlaków sygnałowych powodujących reorganizację mikrofilamentów aktynowych oraz skurcz układu akto-miozynowego. Głównymi regulatorami tych procesów są białka z rodziny Rho, fosfolipidy PIP2 oraz jony wapnia. Receptory nukleotydowe P2Y2 w połączeniu z białkami G regulują poziom fosfatydyloinozytolu-4,5-bisfosforanu (PIP2), który moduluje funkcje białek wiążących aktynę i aktywuje białka Rac1 oraz RhoA. Szlak sygnałowy RhoA/ROCK odgrywa ważną rolę w generowaniu skurczu włókien naprężeniowych. Z kolei białko Rac1 poprzez swój efektor kinazę PAK1 reguluje procesy formujące lamellipodium oraz wysuwanie strefy wiodącej podczas migracji.

Motility is a common feature of numerous cell types. In response to various stimuli, the dynamic actin cytoskeleton and contractility generate forces needed to drive the cell forward. Actin filament elongation on the barbed ends pushes the plasma membrane forward during lamellipodium formation. Stress fibers contraction and/or the contraction of the cortical network are responsible for detaching the rear part of the cell and enable cell body to follow the progressing front. In response to extracellular stimuli, multiple signaling pathways are initiated resulting in the actin filament network reorganization and contractility of acto-myosin system. The key regulators of these processes are Rho family proteins, PIP 2 and calcium ions. Nucleotide receptors P2Y 2 coupled with G-proteins regulate the level of phosphatidylinositol-4,5-bisphosphate (PIP 2 ), which in turn modulates a variety of actin binding proteins, is involved in calcium response, and activates Rac1 and RhoA proteins. The RhoA/ROCK signaling pathway plays an important role in contractile force generation needed for the assembly of stress fibers, focal adhesions and for tail retraction during cell migration. The Rac1 via its effector Pak1 regulates lamellipodium formation and protrusion of the leading edge.

INTRODUCTION In the recent years, the problem of emigration of young people has con- siderably increased, resulting in shortages in the domestic labour market. Aim of the survey was assessment of career plans and reasons for emigra- tion of students of chosen faculties at Polish universities. MATERIAL AND METHODS The survey was conducted at 5 Polish universities at 4 faculties (medicine, dentistry, physiotherapy and cosmetology) with the use of an anonymous author’s questionnaire. RESULTS 40.4% out of the surveyed 1812 individuals are planning to emigrate to other countries after graduation. Their destinations include mainly Great Britain and Scandinavian countries. The students do not specify duration of emigration. In majority of cases they choose the answers “for some time” and “for several years”. No signifi cant diff erences between the facul- ties were found as far as the reason of emigration, its duration and country of destination are concerned. The majority of students are well informed about relevant conditions and are actively preparing themselves to emigra- tion. CONCLUSIONS Over 40% students are planning to emigrate after graduation. The reasons why they emigrate include: salaries, diffi culties in professional education, work organization and professional prestige. Based on the aforementioned reasons for emigration proper solutions, both economic and organization-al ones, should be put in place urgently in order to encourage the young to stay in the country or return from emigration.

WSTĘP Problem emigracji młodych ludzi w ostatnich latach znacznie się nasilił, doprowadzając do niedoborów na krajowym rynku pracy. Celem pracy była ocena planów zawodowych i przyczyn planowanej emigracji studentów wybranych kierunków studiów wyższych w Polsce. MATERIAŁ I METODY Badanie przeprowadzono w 5 polskich uczelniach wyższych na 4 kierunkach (medycyna, stomatologia, fizjoterapia, kosmetologia) przy użyciu anonimowego, autorskiego kwestionariusza. WYNIKI Z przebadanych 1812 studentów 40,4% planuje emigrację po studiach. Kraje docelowe to głównie Wielka Brytania i kraje skandynawskie. Studenci nie precyzują dokładnie czasu trwania emigracji, dominują odpowiedzi „na jakiś czas”, „kilka lat”. Nie stwierdzono istotnych różnic między kierunkami co do przyczyn emigracji, czasu jej trwania i kraju docelowego. Większość studentów jest dobrze poinformowana o warunkach i czynnie przygotowuje się do wyjazdu. WNIOSKI Ponad 40% studentów planuje po ukończeniu studiów emigrację. Przyczyny emigracji to: zarobki, trudności w kształceniu zawodowym, organizacja pracy, prestiż zawodu. Opierając się na wymienionych przyczynach emigracji, należałoby pilnie wdrożyć odpowiednie rozwiązania, zarówno ekonomiczne, jak i organizacyjne w celu zachęcenia młodych ludzi do pozostania w kraju lub powrotu z emigracji.
