Background: 3T3-L1 preadipocyte is the most commonly used cell line in in vivo studies of obesity. One of the main concerns in 3T3-L1 preadipocyte culture is microorganism contaminations. The objective of this study was to determine the appropriate antibiotics to prevent contaminations in 3T3-L1 cultures. Method: This study used descriptive analysis. Frozen 3T3-L1 preadipocytes were thawed and cultured in DMEM-10% FBS-1% penicillin-streptomycin, DMEM-10% FBS-1% penicillin-streptomycin-fungiezone, or DMEM-10% FBS-0.2% ciprofloxacin 200 mg/100 ml. After 24-hour incubation, the cells were observed under the microscope for any change in the medium colour, presence of abnormal structures, and abnormality in cell morphology. Results: The usage of 1% penicillin-streptomycin, 1% penicillin-streptomycin-fungiezone, or 0.2% ciprofloxacin 200 mg/100 ml maintained the clean medium and conserved normal fibroblast-like morphology of the cells. Conclusion: This study suggested that 1% penicillin-streptomycin, 1% penicillin-streptomycin-fungiezone, or 0.2% ciprofloxacin 200 mg/100 ml can be utilized in 3T3-L1 preadipocyte cultures to prevent contaminations.

Water satisfies human needs in many ways. Without water, life is not possible in this planet. According to the estimation of the WHO, 4,000 children die every day from water-borne diseases and this condition is worst in developing and under-developed countries. Water-borne diseases are caused by the consumption of contaminated water. Indeed, it is the main agent of transference of the pathogens causing these diseases. The main objective of this paper is ‘to identify the water-borne diseases and their challenges and to suggest remedial measures to overcome the challenges in the study area’. Both primary and secondary data were used to conduct this study and the analysis of this study is tabulated clearly in the data analysis part. The coastal areas of Ampara District (Sainthamaruthu, Karaitivu, Maligaikadu Mawadipalli, Akkaraipattu, Irakkamam, Varipathanchenai, Addalaichenai, Palamunai, Oluvil and Nintavur) face a lot of challenges induced by water-borne diseases. Viruses, bacteria, parasites and protozoa are the main pathogens. Bacillary dysentery (shigellosis), cholera, other diarrheal diseases, hepatitis A and E and typhoid fever were identified as the most common diseases in these areas. The causes for the diseases are the contamination of water by improper drainage management, effluent contamination of water sources, over population and the lack of awareness among residents about water-borne diseases. According to the result of this study, controlling water contamination, constructing water purification systems, mending existing drainage systems, following instructions when constructing latrines, examining and remediating contaminated wells and providing awareness among residents are ways to attain the objectives.

Radioactive cobalt isotope 60Co is produced in neutron activation process of stable isotope 59Co by neutron capture reaction 59Co(n,g)60Co, in solar wind charge exchange 60Ni(n,p)60Co or in spallation process 62Ni(n,p2n)60Co. These processes are responsible for isotope 60Co production either in nuclear reactor on Earth or beyond – in stars and in meteorites (induced by cosmic rays). In this work the results of 60Co measurements in metal sources by gamma spectrometry laboratory are presented. A metal reference radioactive standards made up of steel cast with discs shaped geometries and different diameters have been tested in gamma-ray spectrometry measurement system. The reference activity concentrations of 60Co were in the range of (0.291±0.010) Bq·g–1 to (1.544±0.030) Bq·g–1. The mean minimal detectable activities (MDA) obtained by series of the 6 to 18 hours lasting measurements of described above standards with HPGe detectors carried out in NCBJ OR POLATOM were in the range of 6.1 mBq·g–1 to 8.5 mBq·g–1. The results correspond to the values of 60Co activity concentration measured in the iron meteorites with young terrestrial ages.

Environmental contamination with heavy metals as a result of anthropogenic activities is not a recent phenomenon. Contaminated sites with heavy metals can be found in functioning as well as abandoned industrial (brownfield) territories, landfills, residential areas with historical contamination, road sides and rarely in polluted sites by natural activities. Pollution data on its amount and concentrations is known from historical studies and monitoring nowadays, but it should be periodically updated for the use of territorial planning or in case of a change of the land use. A special attention should be paid to heavy metal contamination, because in many cases this contamination is most problematic for remediation. 242 territories now are numbered as contaminated and fixed in the National Register of contaminated territories - at least 56 of them are known as contaminated with heavy metals in different amount and concentration. Legislative aspects are discussed as well as an overview of soil and groundwater contamination research and the possible remediation technologies in Latvia are given. Two case studies are described in order to give the inside look in pre-investigations done before potential start of heavy metal remediation works.
