The study aimed to check the effectiveness of anticancer therapy combining a vascular-disruptive drug (combretastatin phosphate, CA4P) and a liposomal formulation of a chemotherapeutic (doxorubicin). CA4P was synthesized in our laboratory according to a previously described procedure. The antivascular drug and long-circulating doxorubicin-loaded liposomes were used to treat B16-F10 murine melanoma experimental tumors. Seventy-four hours after drug administration, a decrease in the number of tumor blood vessels was apparent and necrotic areas within tumors were visible. Combination therapy consisting of alternate administrations of CA4P and liposomal doxorubicin yielded greater inhibition of tumor growth than monotherapies alone. The best therapeutic results were obtained with the antivascular drug administered intratumorally every second day at 50 mg/kg body mass. In the case of combined therapy, the best results were obtained when the vascular-disruptive agent (CA4P) and the antineoplastic agent (liposomal doxorubicin) were administered in alternation.
We investigated suppression of murine B16(F10) melanoma tumor growth following a therapy which involved concomitant administration of cyclophosphamide and plasmid DNA bearing interleukin-12 gene. Since both therapeutic factors display antiangiogenic capabilities, we assumed that their use in blocking the formation of new blood vessels would result in augmented inhibition of tumor growth. This combined therapy regimen indeed resulted in a considerable suppression of tumor growth. We observed a statistically significant extension of treated animals' lifespan. Interestingly, the therapeutic effect was also obtained using a plasmid without an interleukin gene insert. This observation suggests that plasmid DNA, which has been widely applied for treating neoplastic tumors, contains element(s) that elicit immune response in mice.
Rheumatoid arthritis is a common severe joint disease that affects all age groups, it is thus of great importance to develop new strategies for its treatment. The aim of the present study was to examine the combined effect of coenzyme Q10 (CoQ10) and methotrexate (MTX) on the progression of adjuvant-induced arthritis in rats. Adjuvant arthritis (AA) was induced by a single intradermal injection of heat-inactivated Mycobacterium butyricum in incomplete Freund's adjuvant. The experiments included healthy animals, arthritic animals not treated, arthritic animals treated with CoQ10, with methotrexate, and with a combination of CoQ10 and methotrexate. The two latter groups received a daily oral dose of 20 mg/kg b.w. of CoQ10, either alone or with methotrexate in an oral dose of 0.3 mg/kg b.w. twice a week. We found that CoQ10 potentiated both the antiarthritic (decrease of hind paw volume) and the antioxidant effect of methotrexate on the level of oxidation of proteins (suppression of protein carbonyl level in plasma) as well as lipoperoxidation (suppression of levels of HNE-adducts and MDA-adducts to plasma proteins). The same effect was observed for plasmatic levels of CoQ9 and IL-1α, and partially also for γ-glutamyltransferase activity assessed in joints and spleen. Moreover, the combination therapy improved the functionality of peripheral blood neutrophils in AA, with a balancing effect on the immunosuppression caused by MTX monotherapy. In summary, combined administration of CoQ10 and methotrexate suppressed arthritic progression in rats more effectively than did MTX alone. This finding may help improve treatment of rheumatoid arthritis.
Primary arterial hypertension is a long-lasting elevation of blood pressure above the accepted numbers. It is one of the most common causes of mortality in the world. Not until the complications occur it may be symptomless, furthermore it coexists with other cardiovascular risk factors such as metabolic disorders. The aim of medical treatment is to reduce hypertension but also eliminate the risk factors by encouraging changes in the patient’s lifestyle (non-pharmacological actions) and introducing proper therapy. According to Polish Hypertension Society there are five major groups of hypotensive drugs. These are: thiazide diuretics (thiazide-like preferred), betaadrenolytics (vasodilatators preferred), calcium channel antagonists (dihydropiridine preferred), angiotensin converting enzyme inhibitors and angiotensin II receptor blockers. Drugs from the groups above may be used as a monotherapy and also in various specific combination. In the uneventful hypertension the hypotensive therapy should be started from single medication chosen from five main groups of drugs, which have proved favorable influence on the reduction of cardiovascular mortality. But the majority of treated population needs more than one drug to control the hypertension. Then it is necessary to start the combined therapy and use medication in connections with regards to the favorable synergic and additive mechanism of action of each active substance. The remarkable advantages in efficiency, blood vessels protection and avoiding side effects are reached with the combination of angiotensin converting enzyme inhibitors and dihydropirydyne calcium channel antagonist. In the combined therapy it is always necessary to take into account the use of combined pill, because it essentially improves the effectiveness of hypertension treatment and raises the compliance.

Nadciśnienie tętnicze pierwotne to trwałe podwyższenie ciśnienia tętniczego powyżej przyjętych wartości. Jest jedną z najczęstszych przyczyn zgonów na świecie. Przed wystąpieniem powikłań narządowych może nie dawać żadnych objawów, a dodatkowo najczęściej współistnieje z innymi czynnikami ryzyka sercowo-naczyniowego z grupy zaburzeń metabolicznych. Podstawą postępowania medycznego jest normalizacja ciśnienia tętniczego i eliminacja czynników ryzyka, które należy osiągnąć poprzez zmobilizowanie pacjenta do istotnych zmian stylu życia (postępowanie niefarmakologiczne) oraz wdrożenie odpowiedniej farmakoterapii. Zgodnie z wytycznymi Polskiego Towarzystwa Nadciśnienia Tętniczego dysponujemy pięcioma głównymi grupami leków hipotensyjnych. Są to: diuretyki tiazydowe (preferowane tiazydopodobne), beta-adrenolityki (preferowane wazodylatacyjne), antagoniści wapnia (preferowane pochodne dihydropirydynowe), inhibitory enzymu konwertującego angiotensynę oraz blokery receptora AT1 dla angiotensyny drugiej. Leki z tych grupa można stosować w monoterapii, jak również w pewnych wzajemnych połączeniach. W niepowikłanym nadciśnieniu tętniczym terapię hipotensyjną rozpoczyna się od pojedynczego leku, wybierając spośród pięciu głównych grup o udowodnionym korzystnym wpływie na redukcję śmiertelności sercowo-naczyniowej. Większość chorych wymaga jednak stosowania więcej niż jednego leku w celu osiągnięcia docelowych wartości ciśnienia tętniczego. Wówczas należy wdrożyć terapię skojarzoną, stosując leki w połączeniach uwzględniających wypadkowy efekt synergistyczny i addytywny mechanizmów działania poszczególnych substancji czynnych. Szczególnie korzystne pod względem siły działania, pożądanych efektów naczynioprotekcyjnych i minimalizacji działań ubocznych jest połączenie inhibitora konwertazy angiotensyny z antagonistą kanałów wapniowych z grupy pochodnych dihydropirydynowych. W terapii skojarzonej należy zawsze rozważyć stosowanie leków w postaci preparatu złożonego, gdyż wpływa to istotnie na skuteczność leczenia nadciśnienia tętniczego i sprzyja przestrzeganiu zaleceń lekarza.
Pain is integral part of the human life. It is a common disease symptom manifested by most patients visiting clinics. It is a warning signal of a danger or a bodily injury. Occurrence of pain is associated with worse quality of life and negative emotions in the patient. Development of a pain-eliminating therapeutic option is still the biggest challenge to specialists. The purpose of this paper is to evaluate pain control and motor function control in patients taking ketoprofen in the form of prolonged-release capsules with or without applying additionally ketoprofen in the form of gel. The author discusses the efficacy of the pain therapy regime in two therapeutic groups. One group was given combined therapy with the use of ketoprofen in the form of both capsules and gel. The other group was given monotherapy using ketoprofen tablets only. Analysis of obtained results indicates that in both therapeutic groups, the effects of pain in major areas of the subjects’ lives have decreased significantly, their motor functions have improved, and they have gained a better control over pain. Improvement in the abovementioned aspects was however greater in the combined therapy group (capsule + gel). Analysis of the problem is presented basing on a research project conducted by the company Health Data Management in the time from February to July 2011. Data were collected with the use of a questionnaire survey. The study included 3129 patients and 214 physicians.

Ból jest częścią życia ludzkiego, jest nieodłącznie z nim związany. To objaw choroby, z którym zgłasza się do gabinetów lekarskich najwięcej pacjentów. Jest sygnałem ostrzegającym przed zagrożeniem, uszkodzeniem ciała. Z występowaniem bólu wiążą się pogorszenie jakości życia chorego i negatywne emocje. Wypracowanie opcji terapeutycznej niwelującej ból wciąż stanowi największe wyzwanie dla specjalistów. Celem niniejszego artykułu jest ocena kontroli bólu oraz sprawności pacjentów stosujących ketoprofen w postaci kapsułek o przedłużonym uwalnianiu w zależności od dodatkowego stosowania ketoprofenu w postaci żelu. Przeanalizowano skuteczność schematu terapeutycznego w uśmierzaniu bólu w dwóch grupach terapeutycznych. Pierwsza z nich stosowała terapię skojarzoną pod postacią ketoprofenu w kapsułce oraz żelu, zaś druga – monoterapię ketoprofenem w kapsułce. Uzyskane wyniki wskazują, iż w obu grupach terapeutycznych znacznie zmniejszyło się oddziaływanie bólu na główne obszary życia badanych, polepszyła się ich sprawność ruchowa oraz uzyskali oni większą kontrolę nad bólem. Jednakże w grupie, która stosowała terapię skojarzoną (kapsułka + żel), widoczna była większa poprawa w powyższych aspektach. Analiza problemu została przedstawiona w oparciu o wyniki projektu badawczego zrealizowanego przez firmę Health Data Management w okresie od lutego do lipca 2011 roku. W celu zgromadzenia danych posłużono się wywiadem kwestionariuszowym. W badaniu wzięło udział 3129 pacjentów oraz 214 lekarzy
