The study was designed to assess the antioxidant defense mechanisms, either enzymatic or non-enzymatic, in a group of sixteen centenarians (one male and fifteen female subjects aged 101 to 105 years) living in the Upper Silesia district (Poland) in order to evaluate the potential role of antioxidant defenses in human longevity. The results of our preliminary study showed that in comparison with young healthy female adults the centenarians had significantly higher red blood cell glutathione reductase and catalase activities and higher, although insignificantly, serum vitamin E level.

The aim of this study was to evaluate the effects of a regular pre-season training on the aerobic performance and the blood antioxidant defense capacity in soccer players from the Polish Premier League club (n=19) and IVth League team (n=15). The players participated in an incremental treadmill running exercise to volitional fatigue twice (i.e., at the beginning (Trial A) and the end (Trial B) of the pre-season spring training). In venous blood samples, taken at rest and 3 min post-test, the activities of antioxidant enzymes (SOD, GSH-Px, CAT, GR) and concentrations of non-enzymatic antioxidants (GSH, tocopherols, retinol, uric acid) and malondialdehyde as a lipid peroxidation biomarker were measured. With the aim of between-group comparisons and possible conclusions on training-induced changes in the capacity of the blood antioxidant defense, the POTAX index was calculated as a sum of standardized activities of antioxidant enzymes and concentrations of non-enzymatic antioxidants.The results of the present study indicate that the players from the Premier League club were characterized by only slightly higher maximal oxygen uptake rates, the differences compared to IVth League team, as assessed in both trials, were statistically insignificant. Participation in the pre-season training resulted in a moderate improvement of aerobic performance, although only a few players were characterized by VO2max comparable to the international-class elite performers. No distinct differences were observed in the level of aerobic performance between higher- and lower-classified players.Pre-season training led to an improvement in the global blood antioxidant capacity expressed in terms of POTAOX indices, although the changes in the activities and concentrations of individual components of the antioxidant system were less pronounced. Training-induced level of antioxidant conditioning was higher among the Premier League players, which may be related to differences in the training schedule and nutritional preparation of the athletes.

Background: To evaluate whether salivary lead can be used as a surrogate for blood lead, and if so, over what concentration range. Methodology: Three saliva devices were evaluated and one chosen to undertake this project. Paired saliva and blood samples were collected from 89 UK lead workers. Lead concentrations were determined using ICP-MS. In addition, haemoglobin and ZPP levels were determined in the blood samples and albumin was determined in the saliva samples to investigate standardisation using protein adjustments. Results: The chosen saliva device gave low but consistent recoveries for lead in saliva and the blank levels were low. The mean +/- SD blood lead level was 19.9 +/- 14 μg/dl; the mean +/- SD saliva lead level was 19.1 +/- 32.5 μg/l for 89 workers. Log10-transformed data showed correlation of r=0.69. The protein adjustments did not improve the blood-saliva correlation. Conclusions: This study has demonstrated that salivary lead measurement is feasible and correlated with blood lead levels, at least at occupational exposure levels, and may have value as a screening technique. Correlation may improve at environmental levels where exposures are generally more consistent and chronic, although this needs to be demonstrated in a genuine environmental population.

The aim of the present study was to determine the effect of one-day wrestling tournament on magnitude and direction of changes in selected immunological and endocrine status indices. The participants of the study were male (n = 12) and female (n = 13) wrestlers. The earlobe blood samples and unstimulated saliva were collected at three time points: in the morning before the competition (1), immediately after the last match (2) and in the next morning (3). Total protein, IgA and cortisol concentration and α-amylase activity in saliva, as well as concentration of cortisol and interleukin-6 in serum were measured. Significantly lower ratio of sIgA/protein and significantly higher levels of salivary and serum cortisol, interleukin-6, and α-amylase activity were observed at the second time point compared with the first and the third in both groups of athletes. Regardless of the examined time point, concentrations of serum and saliva markers were similar in female and male wrestlers. However, a tendency to lower sAA activity on the next morning after the tournament compared to basal activity of this enzyme was seen in both examined groups. In general, the changes of examined markers were short-lived, except for α-amylase activity, showing that tournament-wrestling matches had no sustained negative effects on endocrine and immunological body systems.

Prion diseases, a group of infectious, fatal neurodegenerations comprise kuru, Creutzfeldt-Jakob disease (CJD), Gerstmann- Sträussler-Scheinker disease (GSS) and fatal familial insomnia (FFI) in man, scrapie in sheep, goats and mouflons, bovine spongiform encephalopathy (BSE) or mad cow disease and chronic wasting disease in elk and mule deer. Variant Creutzfeldt- Jakob disease (vCJD) is a novel human prion disease caused by the bovine spongiform encephalopathy agent. Most cases have occurred in the UK, with smaller numbers in 11 other countries. In Poland, vCJD has not beet observed. All definite vCJD cases have occurred in methionine homozygotes at codon 129 in the prion protein gene (PRNP). Following oral infection, the vCJD agent replicates in lymphoid tissues during the asymptomatic phase of the incubation period. At present, four probable cases of vCJD infection have been identified following transfusion of red blood cells from asymptomatic donors who subsequently died from vCJD. Recently, one case of likely transmission of vCJD infection by UK Factor VIII concentrates has been reported in an elderly haemophilic patient in the UK. The recent report of a blood test that may be used to detect vCJD has raised the possibility of a new way to identify infected individuals, perhaps even before the onset of clinical symptoms.

Choroby wywoływane przez priony obejmują: chorobę Creutzfeldta-Jakoba (Creutzfeldt-Jakob disease, CJD), kuru, chorobę Gerstmanna-Sträusslera-Scheinkera (GSS), śmiertelną rodzinną bezsenność (fatal familial insomnia, FFI) u człowieka, scrapie (polska nieużywana nazwa: trzęsawka) u owiec, kóz i muflonów, pasażowalną encefalopatię norek (transmissible mink encephalopathy, TME), encefalopatię gąbczastą bydła albo chorobę szalonych krów oraz przewlekłą chorobę wyniszczającą u jeleni (chronic wasting disease). Wariant choroby Creutzfeldta-Jakoba (vCJD) jest to nowa choroba z grupy chorób wywoływanych przez priony spowodowana pasażem czynnika encefalopatii gąbczastej bydła (BSE). Przed wystąpieniem vCJD obserwowano pasaż BSE na kota domowego, pumę, ocelota, lwa i bizona. Większość przypadków vCJD obserwowano w Wielkiej Brytanii, mniejszą liczbę w 11 innych krajach. Wszystkie definitywne przypadki vCJD wystąpiły u homozygot Met Met w kodonie 129. genu kodującego PrP (PRNP). W wyniku zakażenia przez przewód pokarmowy czynnik infekcyjny vCJD replikuje się w tkance limfatycznej przed wystąpieniem objawów klinicznych choroby. Obecnie cztery prawdopodobne przypadki vCJD zidentyfikowano u biorców masy czerwonokrwinkowej uzyskanej od asymptomatycznych dawców, którzy następnie zmarli na vCJD. Ostatnio jeden przypadek prawdopodobnego pasażu vCJD poprzez koncentrat czynnika VIII wystąpił w Wielkiej Brytanii u starszego chorego z hemofilią. Ostatnie doniesienie o teście laboratoryjnym wykonywanym we krwi, który może być zastosowany do detekcji vCJD, wskazuje na możliwość identyfikacji zakażonych osób być może jeszcze przed wystąpieniem objawów klinicznych.
