Background: In this work we studied the relationship between the enhanced expression of DR5 receptor and the effect of combination of TRAIL and ionizing radiation on cell cycle arrest and apoptosis induction in human leukemia cell line HL-60. Material and methods: DR5, APO2.7 and cell cycle were analyzed by flow cytometry. Proteins Bid and Mcl-1 were analyzed by Western-blotting. For clonogenic survival, colony assay on methylcellulose was used. Results: Ionizing radiation caused significantly enhanced positivity of DR5 receptors 24 h after irradiation with high doses (6 and 8 Gy). An increase of DR5 receptor positivity after a dose of 2 Gy was not statistically significant and application of TRAIL 48 h after irradiation did not increase the apoptosis induction. However, a decrease of radiation-induced G2 phase arrest and an increase of apoptosis were observed when TRAIL was applied 16 h before irradiation with the dose of 2 Gy. Incubation with 6 µg/l TRAIL for 16 h reduced D0 value from 2.9 Gy to 1.5 Gy. The induction of apoptosis by TRAIL was accompanied by Bid cleavage and a decrease of antiapoptotic Mcl-1 16 h after incubation with TRAIL. Conclusion: TRAIL in concentration of 6 µg/l applied 16 h before irradiation by the dose of 1.5 Gy caused the death of 63% of clonogenic tumor cells, similarly as the dose of 2.9 Gy alone, which is in good correlation with the enhanced apoptosis induction.
Tumor necrosis factor-related apoptosis inducing ligand (TRAIL) is a cytokine of the tumor necrosis factor (TNF) superfamily. The mRNA for TRAIL is expressed in most of the normal human cells and tissues, including T cells, monocytes, macrophages, dentritic cells, natural killer cells (NK) and spleen, lung, prostate. The most important biological function of this cytokine is inducing apoptosis in cancer, transformed cells with little or no cytotoxity against non-transformed cells and tissues and thus TRAIL is promising anticancer cytokine. TRAIL induces programmed cell death through interacting with its receptors. Five TRAIL receptors have been identifi ed: TRAIL-R1 and TRAIL-R2 have ability to initiate the apoptosis- signaling cascade after ligation, whereas “decoy receptors”-TRAILR3, -R4 and osteoprotegerin (OPG) lack this ability. TRAIL induces apoptosis in several tumor cell lines, but many primary tumors are resistant to TRAIL-induced apoptosis. Several mechanisms underlying TRAIL resistance have been proposed. Thus, scientists are currently attempting to identity TRAIL sensitizers that are able to overcome TRAIL resistance in cancer cells. The chemotherapy agents, radiation, lipopolysaccharide, interferons, fl avonoids are capable of enhancing TRAIL-induced apoptosis in cancer cells.

Ligand czynnika martwicy nowotworu (TRAIL) jest cytokiną należącą do nadrodziny czynnika martwicy nowotworów (TNF). Ekspresję mRNA dla TRAIL stwierdzono w wielu prawidłowych komórkach i tkankach organizmu: limfocytach T, monocytach, makrofagach, komórkach dendrytycznych, komórkach natural killer (NK), a także w śledzionie, płucach, gruczole krokowym. Najważniejszą funkcją biologiczną tej cytokiny jest indukowanie apoptozy w komórkach nowotworowych, transformowanych,bez działania cytotoksycznego w stosunku do niezmienionych komórek i tkanek, dlatego TRAIL jest obiecujacą cytokiną antynowotworową. TRAIL indukuje programowaną śmierć komórki poprzez interakcje ze swoimi receptorami. Zidentyfikowano pięć receptorów TRAIL: TRAIL-R1 i TRAIL-R2, mających zdolność inicjowania sygnału do apoptozy po połączeniu z ligandem, oraz „receptory pułapki” – TRAIL-R3 i TRAIL-R4, osteoprotegeryna (OPG) – nieposiadające tej zdolności. TRAIL indukuje apoptozę wielu linii komórek nowotworowych, jednak wiele pierwotnych nowotworów jest opornych na apoptozę indukowaną TRAIL. Zaproponowano wiele mechanizmów leżących u podstaw tej oporności. Naukowcy starają się zidentyfikować czynniki uwrażliwiające komórki nowotworowe w celu przełamania ich oporności na TRAIL. Chemioterapia, promieniowanie, lipopolisacharyd, interferony, flawonoidy są zdolne usprawniania apoptozy indukowanej TRAIL w komórkach nowotworowych.
Prostate cancer is one of the most common malignant tumors in men. Tumor necrosis factor-related apoptosis inducing ligand (TRAIL, Apo-2L) is a cytokine of the tumor necrosis factor (TNF) superfamily. TRAIL possesses the ability to induce apoptosis in prostate cancer cells without toxicity to normal cells, which makes it one of the potential anticancer drugs. TRAIL activates the transduction signal of apoptosis through its interaction with receptors TRAIL-R1/DR4 and TRAIL-R2/DR5 on the surface of prostate cancer cells. The recombined TRAIL and antibodies specific to receptors TRAIL-R1/DR4 and TRAIL-R2/DR5 show satisfactory therapeutic effects in phases I and II of clinical trials. Unfortunately, many tumor cells, including prostate cancer cells appear resistant to TRAIL-mediated apoptosis. The mechanisms of TRAIL-resistance in prostate cancer cells concern many stages of apoptotic pathways. A number of agents, both chemical (cytostatics drugs) as well as physical (gamma radiation), have been identified to sensitize prostate cancer cells and overcome their resistance to TRAIL.

Rak prostaty jest jednym z najczęściej występujących nowotworów złośliwych u mężczyzn. Ligand czynnika martwicy nowotworu indukujący apoptozę (tumor necrosis factor-related apoptosis-inducing ligand – TRAIL, Apo-2L) jest cytokiną należącą do nadrodziny czynnika martwicy nowotworu (tumor necrosis factor – TNF). TRAIL ma zdolność indukowania apoptozy w komórkach raka prostaty nie będąc toksycznym w stosunku do prawidłowych komórek, co czyni go jednym z potencjalnych leków przeciwnowotworowych. Poprzez interakcję z receptorami TRAIL-R1/DR4 i TRAIL-R2/DR5 na powierzchni komórek raka prostaty uruchamia transdukcję sygnału do apoptozy. Rekombinowany TRAIL i przeciwciała swoiste w stosunku do receptorów TRAIL-R1 i TRAIL-R2 wykazują satysfakcjonujące efekty terapeutyczne w badaniach klinicznych I i II fazy. Niestety, wiele komórek nowotworowych, w tym komórek raka prostaty, wykazuje oporność na apoptozę mediowaną przez TRAIL. Mechanizmy TRAIL-oporności komórek raka prostaty dotyczą wielu etapów szlaku apoptozy. Zidentyfikowano szereg czynników chemicznych (cytostatyki) oraz fizycznych (promieniowanie gamma), które mogą uwrażliwiać komórki raka prostaty i przełamać ich oporność na działanie TRAIL.
