The compatibility of the active pharmaceutical ingredient with excipients is required at the drug product development stage. Chemical interactions between the active pharmaceutical ingredient and other components of a dosage form may influence the solubility, bioavailability and pharmacological activity of an active pharmaceutical ingredient and consequently, the efficacy and safety of pharmacotherapy may be changed significantly. In spite of the fact that compatibility testing is recommended by ICH (International Council for Harmonisation of Technical Requirements for Pharmaceuticals for Human Use), there is still a lack of definite and clear guidelines which specify the methodology and conditions of compatibility studies. The aim of this research study was to evaluate the usefulness of Attenuated Total Reflectance Fourier-Transform Infrared Spectroscopy technique (ATR-FTIR) in monitoring changes in gabapentin structure as a result of chemical interactions with selected excipients. The samples of binary mixtures containing equal masses of gabapentin and selected excipients were analyzed. Infrared spectra were recorded in the range of wavenumbers from 4000 cm-1 to 600 cm-1 immediately after the samples preparation and after 5 days of their exposure to the elevated temperature of 50°C. The analysis of registered ATR-FTIR spectra indicated some shifts in absorption bands, modifications of their shape or intensity, and the disappearance or appearance of additional peaks. These changes in infrared spectra confirmed the occurrence of chemical interactions between gabapentin and selected excipients. The usefulness of ATR-FTIR spectroscopy as a rapid screening technique for preliminary analysis of chemical interactions of gabapentin has been shown. For a thorough evaluation of the chemical interactions, further studies are recommended by means of complementary techniques such as Raman spectroscopy, differential scanning calorimetry or high-performance liquid chromatography.

Ocena zgodności chemicznej substancji czynnej (ang. Active Pharmaceutical Ingredient, API) z substancjami pomocniczymi jest jednym z podstawowych badań na etapie opracowywania nowej formulacji farmaceutycznej. Interakcje pomiędzy API a pozostałymi składnikami postaci leku mogą istotnie wpływać m.in. na rozpuszczalność, biodostępność oraz właściwości farmakologiczne substancji czynnej, a w konsekwencji rzutować na skuteczność i bezpieczeństwo farmakoterapii. Pomimo wymogów przeprowadzania badań niezgodności, wciąż brakuje konkretnych wytycznych precyzujących metodologię oraz warunki wykonywania tego typu analiz. Celem pracy była ocena przydatności metody spektroskopii osłabionego całkowitego odbicia w zakresie podczerwieni (ang. Attenuated Total Reflactance Spectroscopy, ATR-FTIR) w monitorowaniu zmian zachodzących w strukturze cząsteczki gabapentyny w wyniku interakcji chemicznych z substancjami pomocniczymi. Analizie poddano próbki równowagowych, dwuskładnikowych mieszanin gabapentyny z wybranymi substancjami pomocniczymi. Widma IR zarejestrowano w zakresie liczb falowych od 4000 cm-1 do 600 cm-1 bezpośrednio po wykonaniu próbek, a także po ich 5-dniowej ekspozycji na temperaturę 50°C. Zarejestrowana zmiana położenia i intensywności pasm absorpcji, zanik pasm lub pojawienie się nowych pasm absorpcji w widmie IR wskazuje na wystąpienie interakcji chemicznych między gabapentyną a wybranymi substancjami pomocniczymi. Wykazano użyteczność metody ATR-FTIR jako szybkiej nieinwazyjnej techniki przesiewowej, przydatnej we wstępnej ocenie interakcji chemicznych gabapentyny z substancjami pomocniczymi. W celu pełnej oceny interakcji chemicznych wskazane byłoby przeprowadzenie dalszych badań z zastosowaniem technik komplementarnych, jak np. spektroskopia ramanowska, analiza chromatograficzna czy różnicowa kalorymetria skaningowa.
An air diffuse coplanar surface barrier discharge is used to activate the surface of polytetrafluoroethylene (PTFE) samples, which are subsequently coated with polyvinylpyrrolidone (PVP) and tannic acid (TAN) single, bi- and multilayers, respectively, using the dip-coating method. The surfaces are characterized by X-ray Photoelectron Spectroscopy (XPS), Attenuated Total Reflection – Fourier Transform Infrared Spectroscopy (ATR-FTIR) and Atomic Force Microscopy (AFM). The XPS measurements show that with plasma treatment the F/C atomic ratio in the PTFE surface decreases, due to the diminution of the concentration of CF2 moieties, and also oxygen incorporation through formation of new C–O, C=O and O=C–O bonds can be observed. In the case of coated samples, the new bonds indicated by XPS show the bonding between the organic layer and the surface, and thus the stability of layers, while the gradual decrease of the concentration of F atoms with the number of deposited layers proves the creation of PVP/TAN bi- and multi-layers. According to the ATR-FTIR spectra, in the case of PVP/TAN multilayer hydrogen bonding develops between the PVP and TAN, which assures the stability of the multilayer. The AFM lateral friction measurements show that the macromolecular layers homogeneously coat the plasma treated PTFE surface.
