Acute pancreatitis (AP) is a potentially fatal disease. In animal experiments leptin and ghrelin were shown to modulate the course of AP. The aim of the study was to estimate the relationship between the severity of acute biliary pancreatitis (ABP) and serum levels of leptin and ghrelin in nonobese patients in the first seven days of the hospitalization. Material and methods. The study included nine patients with mild ABP (MABP), eleven patients with severe ABP (SABP) and twenty healthy controls, appropriately matched age, sex and weight. Serum concentrations of leptin and ghrelin were measured in patients on the first, third, fifth, and seventh days of hospitalization using leptin and ghrelin RadioImmunoAssay (RIA) kits. Results. At admission and throughout the study the mean serum leptin concentration in SABP patients was higher than in the controls but without statistical significance. Serum ghrelin concentrations on admission were significantly lower in patients with ABP than in the controls. We observed steadily increasing serum ghrelin levels in both groups of the patients during the course of ABP. Conclusions. The results of our study do not support the role of leptin as a marker of the severity of ABP. On the other hand, rising serum ghrelin levels during the course of ABP may be a marker of recovery and an indicator of the healing process

Apart from Na+,K+-ATPase, a second sodium pump, Na+-stimulated, K+-independent ATPase (Na+-ATPase) is expressed in proximal convoluted tubule of the mammalian kidney. The aim of this study was to develop a method of Na+-ATPase assay based on the method previously used by us to measure Na+,K+-ATPase activity (Acta Biochim Polon.; 2002, 49: 515-27). The ATPase activity was assayed as the amount of inorganic phosphate liberated from ATP by isolated microsomal fraction. Na+-ATPase activity was calculated as the difference between the activities measured in the presence and in the absence of 50 mM NaCl. Na+-ATPase activity was detected in the renal cortex (3.5 ± 0.2 μmol phosphate/h per mg protein), but not in the renal medulla. Na+-ATPase was not inhibited by ouabain or an H+,K+-ATPase inhibitor, Sch 28080, but was almost completely blocked by 2 mM furosemide. Leptin administered intraperitoneally (1 mg/kg) decreased the Na+,K+-ATPase activity in the renal medulla at 0.5 and 1 h by 22.1% and 27.1%, respectively, but had no effect on Na+-ATPase in the renal cortex. Chronic hyperleptinemia induced by repeated subcutaneous leptin injections (0.25 mg/kg twice daily for 7 days) increased cortical Na+,K+-ATPase, medullary Na+,K+-ATPase and cortical Na+-ATPase by 32.4%, 84.2% and 62.9%, respectively. In rats with dietary-induced obesity, the Na+,K+- ATPase activity was higher in the renal cortex and medulla by 19.7% and 34.3%, respectively, but Na+-ATPase was not different from control. These data indicate that both renal Na+-dependent ATPases are separately regulated and that up-regulation of Na+-ATPase may contribute to Na+ retention and arterial hypertension induced by chronic hyperleptinemia.

With the increasing importance of early type 2 diabetes (DM2) and obesity detection, it is useful to reevaluate leptin role in these conditions. Our study aimed at investigating circulating leptin concentrations in a group of patients with DM2, and at assessing in detail whether leptin concentrations correlate with selected biochemical, clinical parameters and markers of systemic inflammation in patients with DM2 and in healthy volunteers. In our work, we analysed samples and data drawn from 71 patients aged 61.4 ± 11.7 years, who have been diagnosed with type 2 diabetes, as well as from a healthy control group (HC) consisting of 51 healthy subjects with a mean age of 57.8 ± 13.7 years. Therein, the concentration of leptin in the DM2 patients was significantly higher than in the HC (p < 0.01), with median value of 16.59 (IQR 8.58-33.39) ng/ml in the DM2, vs median value of 6.66 (IQR 4.52-21.40) ng/ml in the HC. In the analysis of variance, higher leptin concentrations were revealed in the DM2 group as compared to the HC, and this figure remained significant after adjusting for gender and age (p < 0.001). Moreover, it was independent of HOMA-IR (p = 0.003). However, the differences in leptin levels between the groups disappeared when additional adjustments for anthropometric parameters (BMI, waist circumference) were applied (p = 0.088). Beyond the aforementioned, significant positive correlations were found in the DM 2 group between leptin level and CRP (r=0.256; p < 0.05) and IL-6 (r = 0.345; p < 0.01). Among the selected variables, only gender and BMI were included in the predictive model explaining the variability of leptin, and, in total, were responsible for 72.6% of the original variation of the studied adipocytokine. The results of this study have led to conclusion that leptin may participate in the complex pathogenesis of DM2 and be a predictor of the development of this disease. As higher concentrations of leptin coexist with obesity, and this situation correlates positively with markers of inflammation (CRP, IL-6), leptin level, hence, should be considered in the pathogenesis of DM2.

Cardiovascular risk of prediabetes is still subject to controversies. We analyzed the associations between insulin resistance, adipokines and incipient atherosclerosis estimated by intima-media thickness (IMT) in a cross-sectional study on 122 prediabetic subjects without clinical signs of atherosclerotic disease. Homeostasis model assessment of insulin resistance (HOMA-IR, calculated as fasting insulin × fasting plasma glucose / 22.5), adiponectin, leptin, leptin-to-adiponectin ratio, carotid and femoral IMT were evaluated. We also assessed other parameters related to insulin resistance and adipokines (HbA1c, anthropometric and lipid parameters), as they may also influence atherosclerosis. Carotid IMT was correlated to adiponectin and leptin-to-adiponectin ratio (all p < 0.05), but not with HOMA-IR or leptin, while femoral IMT showed no relationship with these factors. After adjusting for leptin, leptin-to-adiponectin ratio, triglycerides, HDL-cholesterol, cholesterol-to-HDL ratio, triglycerides-to-HDL ratio and HbA1c, IMT values became correlated with HOMA-IR. Adjustment for HOMA-IR induced the appearance of new correlations between adipokines and both IMT values. In conclusion, insulin resistance and adipokines seem related to IMT in prediabetic subjects without clinical signs of arterial obstruction.

The study aimed to examine the combined effects of caffeine and aerobic exercise on leptin levels and some indices of insulin resistance in diabetics Thirty-two males with type 2 diabetes participated in a quasi-experimental and double-blind design. All participants were divided into four homogeneous groups of 8 individuals, including placebo (P), caffeine supplementation (C), aerobic training (AT), and aerobic training and caffeine (AT + C). The design protocol included eight weeks of aerobic exercise and caffeine consumption. Blood samples were taken to measure serum levels of leptin, glucose, insulin, HbA1c, HOMA-IR, and insulin sensitivity (QUICKI) indices at two phases. Data were analyzed by repeated measure ANOVA, Bonferroni posthoc, and independent T-test at a significant level of a ≤ 0.05. The results showed that the levels of leptin, glucose, insulin, HbA1c, and HOMA-IR in the three intervention groups significantly decreased compared to the placebo group (P = 0.001). In addition, QUICKI was significantly increased in the three groups of caffeine (C), aerobic training (AT), and aerobic training + caffeine (AT + C) compared to the placebo group (P = 0.001). Also, the AT+C group has double effects on the investigated indices compared to the caffeine (C) group (P = 0.001). Regular aerobic exercise and caffeine supplementation may be more effective treatments for improving insulin resistance indicators associated with type 2 diabetes.

Bariatric surgery is the most effective method to achieve weight loss in obese subjects. The aim of this study was to evaluate some adipocytokines and insulin, as well as parameters of metabolic syndrome of the obese patients, for three and six months after vertical banded gastroplasty, in the time of dynamic weight loss. Seven males and two females aged 28 to 49 years, with long lasting simple obesity and the presence of metabolic syndrome, were studied. After surgical treatment the values of the body mass index, waist circumference, systolic and diastolic blood pressure, total cholesterol, LDL cholesterol, triglycerides, and blood concentrations of leptin decreased significantly. Before surgical operation of all obese patients no statistically significant correlations between studied parameters were noted. Three and six months later a lot of correlations between studied parameters appeared. In conclusion, (a) vertical-banded gastroplasty is a valuable method in treatment of obese subjects, leading to a significant decrease in body weight and improvement in some parameters of metabolic syndrome in a few months after surgery, (b) adipocytokines, together with an unknown gastric factor, may be key factors in the control of some features of the metabolic syndrome.

The aim of this study was to identify SNPs in leptin (LEP), leptin receptor (LEPR) and growth hormone (GH) genes in order to analyze genetic diversity of Slovak Spotted cattle. The total numbers of blood samples were taken from 353 Slovak Spotted cows originating from four farms. Genomic DNA was isolated by phenol-chloroform extraction method and analyzed by PCR-RFLP method. After digestion with restriction, enzymes were detected in whole population of cow's alleles with frequency: LEP/Sau3AI A 0.84 and B 0.16 (±0.0152); LEPR/BseGI C 0.95 and T 0.05 (±0.0089) and GH/AluI L 0.70 and V 0.30 (±0.0188). Based on the observed vs. expected genotypes frequencies populations across loci were in Hardy-Weinberg equilibrium (P\>0.05). Predominant for SNP LEP/Sau3AI was AA genotype (0.70), for SNP LEPR/T945M CC genotype (0.91), and LL genotype (0.48) was most frequent for SNP GH/AluI. The observed heterozygosity of SNPs across populations was also transferred to the low or median polymorphic information content 0.24 (He 0.28), 0.08 (He 0.09) and 0.33 (He 0.47) for LEP, LEPR and GH genes, respectively. Within genetic variability estimating negative values of fixation indexes FIS (-0.09-0.05) and FIT (-0.07-0.03) indicating heterozygote excess were observed. The value of FST indexes (0.018-0.023) shows very low levels of genetic differentiation in allele frequencies of loci among evaluated subpopulations. The low values of genetic distances (0.0018-0.0159) indicated high genetic relatedness among animals in subpopulations caused probably by common ancestry used in breeding program at farms.

Leptin and leptin receptor genes are considered as production traits markers in dairy or beef cattle. The aim of this study was to verify the associations of polymorphisms in bovine LEP and LEPR genes with production and reproduction traits in Slovak Spotted and Pinzgau cows. Long-life production was evaluated: milk, protein, and fat yield and reproduction traits: age at first calving, calving interval, days open, and insemination interval. In total, 296 blood samples of Slovak Spotted and 85 hair roots samples of Pinzgau cows were analyzed. In order to detect LEP/Sau3AI (BTA 4, inron 2) and LEPR/T945M (BTA 3, exon 20) genotypes PCR-RFLP method was used. In Slovak Spotted and Pinzgau cows allele frequencies were 0.838/0.162 and 0.694/0.306 for A and B LEP variants, and 0.954/0.046 and 0.912/0.088 for C and T LEPR variants, respectively. For testing the associations between SNPs LEP/Sau3AI and LEPR/T945M and evaluated traits, the General Linear Model procedure in SAS Software was used. Statistical analysis showed that SNP LEP/Sau3AI significantly affected milk, protein and fat yield (P<0.05), and age at first calving (P<0.01) in analyzed population of cows. Statistically, SNP LEPR/T945M affected significantly calving interval (P<0.01) only. Results of our study suggest that especially leptin is a candidate gene, which influences mainly milk production traits and might be implemented in breeding strategies to improve the production performance of both analyzed cattle breeds.

Introduction: Obesity is a syndrome characterized by weight gain over optimal weight which affects both adults and children. The aim of the study was to search for the relation between anthropometric parameters, lipid parameters, glucose, leptin and ghrelin concentrations in patients before and after the implantation of balloon–type gastric BIB (ORBERA). Material and methods: The material for the study was the whole blood derived from 45 people, including 35 women and 10 men voluntarily undergoing gastric balloon implantation. The concentration of glucose, total cholesterol, HDL and LDL, triglycerides, glucose, leptin and ghrelin were measured both before the implantation of the gastric balloon and after its removal. Simultaneously, the evaluation of anthropometric parameters such as body mass index (BMI), body fat distribution (WHR),% body fat and the excess weight loss/weight loss ratio EWL/WL) was done. Results: The statistical analysis showed a significant change of all measured parameters after gastric balloon implantation. There was a statistically significant decrease in variables such as body mass index (BMI), waist–to–hip ratio (WHR), % of body fat, excess weight loss/weight loss (ELW/WL), blood the concentration of glucose, cholesterol (total and LDL), triglycerides and leptin. Concentrations of HDL cholesterol and ghrelin increased significantly. Conclusion: The implantation of a balloon–type gastric BIB (ORBERA) has a beneficial effect on lipid parameters, glucose, leptin and ghrelin concentration. Moreover, it is connected with an the improvement of studied anthropometric parameters.

Wstęp: Otyłość jest zespołem chorobowym cechującym się m.in. zwiększeniem masy ciała powyżej wartości optymalnej i dotyczy zarówno osób dorosłych, jak i dzieci. Celem pracy było poszukiwanie zależności między wskaźnikami antropometrycznymi, parametrami lipidogramu, stężeniami glukozy, leptyny oraz greliny u osób przed i po implantacji balonu żołądkowego typu BIB (ORBERA). Materiały i metody: Materiał do badania stanowiła krew pełna pochodząca od 45 osób, w tym 35 kobiet i 10 mężczyzn, dobrowolnie poddających się implantacji balonu żołądkowego. Zbadano stężenie glukozy, cholesterolu całkowitego, frakcji HDL i LDL, triglicerydów oraz stężenie leptyny i greliny oraz dokonano oceny parametrów antropometrycznych: wskaźnika masy ciała (BMI), wskaźnika dystrybucji tkanki tłuszczowej (WHR), % tkanki tłuszczowej oraz wskaźnika EWL/WL (ang. excess weight loss/weight loss) zarówno przed implantacją balonu żołądkowego, jak i po jego usunięciu. Wyniki: Analiza statystyczna wykazała istotną zmianę wszystkich mierzonych parametrów po implantacji balonu żołądkowego. Stwierdzono statystycznie istotny spadek wartości takich zmiennych, jak: BMI, WHR, % tkanki tłuszczowej, ELW/WL, stężenie glukozy, cholesterolu, triglicerydów, cholesterolu frakcji LDL i leptyny. Wartości stężenia cholesterolu frakcji HDL i greliny istotnie wzrosły. Wnioski: Zabieg implantacji balonu żołądkowego typu BIB (ORBERA) wpływa korzystnie na parametry gospodarki lipidowej, stężenia glukozy, greliny i leptyny, jak również wiąże się z poprawą badanych parametrów antropometrycznych.

The aim of this work was to develop a method for renal H+,K+-ATPase measurement based on the previously used Na+,K+-ATPase assay (Bełtowski et al.: J Physiol Pharmacol.; 1998, 49: 625-37). ATPase activity was assessed by measuring the amount of inorganic phosphate liberated from ATP by isolated microsomal fraction. Both ouabain-sensitive and ouabain-resistant K+-stimulated and Na+-independent ATPase activity was detected in the renal cortex and medulla. These activities were blocked by 0.2 mM imidazolpyridine derivative, Sch 28080. The method for ouabain- sensitive H+,K+-ATPase assay is characterized by good reproducibility, linearity and recovery. In contrast, the assay for ouabain-resistant H+,K+-ATPase was unsatisfactory, probably due to low activity of this enzyme. Ouabain-sensitive H+,K+-ATPase was stimulated by K+ with Km of 0.26 ± 0.04 mM and 0.69 ± 0.11 mM in cortex and medulla, respectively, and was inhibited by ouabain (Ki of 2.9 ± 0.3 μM in the renal cortex and 1.9 ± 0.4 μM in the renal medulla) and by Sch 28080 (Ki of 1.8 ± 0.5 μM and 2.5 ± 0.9 μM in cortex and medulla, respectively). We found that ouabain-sensitive H+,K+-ATPase accounted for about 12% of total ouabain-sensitive activity in the Na+,K+-ATPase assay. Therefore, we suggest to use Sch 28080 during Na+,K+-ATPase measurement to block H+,K+-ATPase and improve the assay specificity. Leptin administered intraperitoneally (1 mg/kg) decreased renal medullary Na+,K+-ATPase activity by 32.1% at 1 h after injection but had no effect on H+,K+-ATPase activity suggesting that the two renal ouabain-sensitive ATPases are separately regulated.

The adipokines produced in the adipose tissue contribute to the etiopathogenesis of arterial hypertension, lipid disorders, atherosclerosis, ischaemic heart disease or insulin resistance. Leptin is one of the adipokines involved in the development of these diseases. The aim of our study was quantitative leptins assessment in the blood serum of patients with systemic sclerosis. Furthermore, we evaluated the correlations between: leptin blood concentration and duration of the disease, the erythrocyte sedimentation rate as well as the value of Rodnan skin score. Quantitative assessment of leptin concentration was carried out in the blood serum of patients with systemic sclerosis and healthy people, with the use of immunoenzymatic method. The research has proved that leptin level in blood serum of systemic sclerosis patients is decreased. Also, we have confi rmed the existence of clear correlation between leptinemy and the values of duration of the disease, the erythrocyte sedimentation rate as well as the value of Rodnan skin score.

Wytwarzane w tkance tłuszczowej substancje, tzw. adipokiny, uczestniczą w etiopatogenezie nadciśnienia tętniczego, zaburzeń lipidowych, miażdżycy, choroby niedokrwiennej serca oraz insulinooporności. Jedną z adipokin uczestniczących w rozwoju wymienionych schorzeń jest leptyna, której ilościową ocenę w surowicy krwi osób chorych na twardzinę układową przyjęto za cel niniejszej pracy. Dla realizacji nadrzędnego celu postanowiono także ocenić zależność między stężeniem tego hormonu w surowicy krwi a czasem trwania choroby, wartością OB oraz wielkością indeksu Rodnana. Ilościowej oceny stężenia leptyny w surowicy krwi osób chorych na twardzinę układową oraz osób zdrowych dokonano metodą immunoenzymatyczną. Stwierdzono obniżenie stężenia leptyny w surowicy krwi osób chorych na twardzinę układową. Ponadto dowiedziono, że istnieje wyraźny związek między leptynemią a czasem trwania choroby, wartością OB oraz wielkością indeksu Rodnana.

Leptin, secreted by adipose tissue, is involved in the pathogenesis of arterial hypertension, however, the mechanisms through which leptin increases blood pressure are incompletely elucidated. We investigated the effect of leptin, administered for different time periods, on renal Na+,K+-ATPase activity in the rat. Leptin was infused under anesthesia into the abdominal aorta proximally to the renal arteries for 0.5-3 h. Leptin administered at doses of 1 and 10 µg/min per kg for 30 min decreased the Na+,K+-ATPase activity in the renal medulla. This effect disappeared when the hormone was infused for ≥1 h. Leptin infused for 3 h increased the Na+,K+-ATPase activity in the renal cortex and medulla. The stimulatory effect was abolished by a specific inhibitor of Janus kinases (JAKs), tyrphostin AG490, as well as by an NAD(P)H oxidase inhibitor, apocynin. Leptin increased urinary excretion of hydrogen peroxide (H2O2) between 2 and 3 h of infusion. The effect of leptin on renal Na+,K+-ATPase and urinary H2O2 was augmented by a superoxide dismutase mimetic, tempol, and was abolished by catalase. In addition, infusion of H2O2 for 30 min increased the Na+,K+-ATPase activity. Inhibitors of extracellular signal regulated kinases (ERKs), PD98059 or U0126, prevented Na+,K+-ATPase stimulation by leptin and H2O2. These data indicate that leptin, by acting directly within the kidney, has a delayed stimulatory effect on Na+,K+-ATPase, mediated by JAKs, H2O2 and ERKs. This mechanism may contribute to the abnormal renal Na+ handling in diseases associated with chronic hyperleptinemia such as diabetes and obesity.

The aim of the research was to assess changes in ghrelin, leptin and free fatty acids (FFA) levels in women’s blood after training. The research was carried out in women aged 45.55 ±11.33 years and with the BMI of 26.49 ±4.49. Health training at 50–66% VO2max took place twice a week for 9 months. In the baseline phase and in the 3rd, 6th and 9th month of the training, body mass and composition were measured, cardiorespiratory fitness was checked after a 10-minute exercise test on a cycloergometer, and fasting levels of ghrelin, leptin and FFA in the serum were assayed and 15 minutes after the exercise test. Body mass was reduced in the 6th month of the training. Fasting ghrelin level increased because of training, leptin and FFA decreased. After single 10-minute exercises performed every 3 months level of ghrelin and FFA increased while leptin decreased. An increase in ghrelin level in the blood after the single exercise can be the result of negative energy expenditure. An increase in fasting ghrelin level after training can be one of the adaptive physiological mechanisms connected with energy saving. A mechanism that is switched on as a result of a long-lasting stimulus that leads to energy losses, reduction in body mass and a decrease in leptin level in the blood.

Abdominal obesity becomes very significant health’s problem, especially because it is connected with pathogenesis of cardiovascular diseases. Adipose tissue is not only a store of excess energy but a hormonally active system too. The substances produced by adipose tissue are adipocytokines. Two of them are leptin and adiponectin. Adiponectin levels are inversely related to the adiposity degree, despite of adipose tissue is only source of it. concentrations of adiponectin have been reported to be decreased in patients with coronary artery diseases, type II diabetes mellitus, hypertensions and dyslipidemia patients in some insulin resistant states. It takes part in processes regulate glucose and lipid metabolism and it has anti-inflammatory and antiatherogenic properties. Adiponectin has a potential protective ability towards to cardiovascular diseases. Positive correlation with degree of adiposity has been reported for leptin – hormone involved in the regulation of food intake and energy expenditure. Leptin exerts many potentially atherogenic effects. It has been reported to influence on arterial hypertension, endothelial dysfunction, platelet aggregation, insulin resistant and activation of sympathetic system. In this way it can play very important role in development of stroke. Recent studies suggest that adiponectin and leptin may play an important role in obesity-associated cerebrovascular diseases. There is still too little evidence to say that these two hormones are independent marks of ischemic stroke and confirm their role in stroke pathogenesis.

Otyłość brzuszna staje się coraz istotniejszym problemem zdrowotnym, głównie ze względu na związaną z nią koincydencję rozwoju chorób sercowo-naczyniowych. Tkanka tłuszczowa jest nie tylko źródłem zapasów energii, ale również hormonalnie czynnym systemem. Substancje wydzielane przez komórki tkanki tłuszczowej to adipocytokiny, do których zaliczamy między innymi leptynę i adiponektynę. Pomimo iż tkanka tłuszczowa jest jedynym źródłem adiponektyny, paradoksalnie jej stężenie w otyłości ulega obniżeniu. Ponadto niższe stężenia wykazano u chorych z chorobą niedokrwienną serca, cukrzycą typu II, nadciśnieniem tętniczym i dyslipidemią, czyli w stanach prawdopodobnej insulinooporności. Adiponektyna bierze udział w procesach usprawniających metabolizm glukozy i lipidów oraz wykazuje działanie przeciwzapalne i przeciwmiażdżycowe, a więc i potencjalnie ochronne działanie wobec chorób sercowo-naczyniowych. Ze wzrostem zgromadzonej tkanki tłuszczowej pozytywnie koreluje poziom leptyny, hormonu uczestniczącego w regulacji poboru pożywienia i wykorzystania energii. Leptyna wykazuje wiele potencjalnie proaterogennych właściwości. Poprzez między innymi wpływ na ciśnienie krwi, układ fibrynolityczny, funkcje śródbłonka, agregację płytek, oporność na insulinę oraz aktywację układu sympatycznego może odgrywać istotną rolę w rozwoju niedokrwiennego udaru mózgu. Badania sugerują, że adiponektyna i leptyna mogą stanowić ogniwo łączące otyłość z chorobami naczyniowymi OUN. Wciąż jednak istnieje zbyt mało danych, by uznać oba hormony za niezależne markery wystąpienia udaru niedokrwiennego mózgu oraz potwierdzić ich rolę w jego patogenezie.

INTRODUCTION: Overweight and obesity have become serious public health problems. They are associated with leptin and the leptin receptor (LEPR). LEPR is one of the gp130 family proteins, which plays a great role in the human body. A number of studies have evaluated many LEPR polymorphisms. MATERIAL AND METHODS: The study included a group of 510 patients residing in the Upper Silesian Agglomeration (divided into healthy, overweight and obesity groups). The LEPR gene polymorphism was investigated using the Applied Biosystems 7300 Real-Time PCR System. The analysis was carried out with fluorescent-labeled probes by means of ready-to-use assay kits for single nucleotide polymorphism detection. RESULTS: There were no statistically significant differences in the distribution of genotypes in the healthy and overweight and obese subjects either in the whole group or in the male and female groups (p > 0.05). The incidence of allele A was 76.73% and allele G 23.27%. CONCLUSIONS: There were no significant differences in the distribution of the rs1137100 LEPR polymorphism for the development of overweight and obesity pathogenesis.

WSTĘP: Nadwaga i otyłość stały się w obecnych czasach poważnym problemem zdrowia publicznego. Patogenezę tych schorzeń można wiązać m.in. z hormonem tkankowym – leptyną, a także jej receptorem kodowanym przez gen LEPR. Receptor leptyny, należący do rodziny białek gp130, odgrywa ogromną rolę w ludzkim organizmie m.in. w patogenezie nadwagi i otyłości. MATERIAŁ I METODY: Badaniem objęto 510 osób. Grupę badaną podzielono na trzy grupy: kontrolną – zdrowych, z nadwagą oraz otyłością. Polimorfizm genu dla receptora leptyny K109R (rs1137100) zbadano w reakcji łańcuchowej polimerazy za pomocą aparatu 7300 Real Time PCR System (Applied Biosystems). Do genotypowania wykorzystano komplementarne do badanych alleli, fluorescencyjnie znakowane sondy TaqMan Predesigned SNP Genotyping Assay (Applied Biosystems). WYNIKI: Nie wykazano znamiennych statystycznie różnic w rozkładzie genotypów między pacjentami zdrowymi a badanymi z nadwagą i z otyłością zarówno w całej grupie, jak i w grupach kobiet i mężczyzn (p > 0,05). Częstość występowania allelu A wynosiła 76,73%, allelu G 23,27%. WNIOSKI: Brak znamiennych różnic w rozkładzie polimorfizmu rs1137100 LEPR w patogenezie nadwagi i otyłości.

INTRODUCTION: Overweight and obesity have become serious public health problems. They are associated with leptin and the leptin receptor (LEPR). LEPR is one of the gp130 family proteins, which plays a great role in the human body. A number of studies have evaluated many LEPR polymorphisms. MATERIAL AND METHODS: The study included a group of 510 patients residing in the Upper Silesian Agglomeration (divided into healthy, overweight and obesity groups). INTRODUCTION: Leptin administered intracerebroventricularly (icv) evokes the resuscitating effect in haemorrhage-shocked rats, and the action is associated with activation of the histaminergic system. The aim of the present study was to examine the possible role of the histamine H3 receptor in leptin-evoked cardiovascular effects. MATERIALS AND METHODS: Studies were performed in male Wistar rats subjected to irreversible haemorrhagic shock (0% survival at 2 h) with a mean arterial pressure (MAP) of 20–25 mmHg that were anaesthetized with keta-mine/xylazine (100 mg/kg + 10 mg/kg, intraperitoneally). At 5 minutes of critical hypotension the animals were injec-ted icv with leptin (20 μg) or 0.9% NaCl (5 μl). RESULTS: Haemorrhage resulted in a decrease in pulse pressure (PP) and heart rate (HR). Leptin evoked long-lasting rises in MAP, PP and HR with 100% survival at 2 h. Pre-treatment with histamine H3 receptor inverse agonist/H4 partial agonist clobenpropit (42.5 nmol, icv), but not with H3 receptor neutral antagonist VUF 5681 (50 nmol, icv), inhibited MAP and PP changes evoked by leptin, however, without an influence on HR or survival rate at 2 h. CONCLUSIONS: The histamine H3 receptor can influence the centrally-acting leptin-induced resuscitating effect in haemorrhagic shock in rats.

WSTĘP: Leptyna podawana do komory bocznej mózgu (icv) wywołuje efekt resuscytacyjny u szczurów we wstrząsie krwotocznym; w działaniu tym uczestniczy układ histaminergiczny. Celem obecnej pracy było zbadanie możliwego udziału receptorów histaminowych H3 w wywoływanym przez leptynę wpływie na układ krążenia we wstrząsie. MATERIAŁ I METODY: Badania przeprowadzono u szczurów, samców szczepu Wistar w znieczuleniu ogólnym przy użyciu ketaminy i ksylazyny (100 mg/kg + 10 mg/kg dootrzewnowo), u których wywołano nieodwracalny wstrząs krwotoczny ze średnim ciśnieniem tętniczym (MAP) 20–25 mmHg (wskaźnik przeżycia dwugodzinnego: 0%). W 5 min krytycznej hipotensji zwierzętom podawano icv leptynę (20 μg) bądź 0,9% roztwór NaCl (5 μl). WYNIKI: Utrata krwi prowadziła do obniżenia ciśnienia tętna (PP) i częstości rytmu serca (HR). Leptyna wywoływała długotrwałe wzrosty MAP, PP i HR, a także podwyższenie do 100% wskaźnika przeżycia dwugodzinnego. Premedykacja odwrotnym agonistą receptorów H3/częściowym agonistą receptorów H4 klobenpropitem (42,5 nmol, icv), ale nie neutralnym antagonistą receptorów H3 VUF 5681 (50 nmol, icv), hamowała zmiany MAP i PP wywoływane przez leptynę, jednak bez wpływu na wskaźnik przeżycia dwugodzinnego we wstrząsie. WNIOSKI: Receptory histaminowe H3 mogą wpływać na efekt resuscytacyjny wywoływany przez ośrodkowo działającą leptynę we wstrząsie krwotocznym u szczurów.
