A homozygous population derived from hybrids between two homozygous parents may be used for genetic analysis of metrical traits. The paper describes the use of doubled haploids (DH) and single seed descent (SSD) lines for detection of linkage between genes conditioning two quantitative traits. A computational algorithm is presented, which facilitates matching various variants of relations between variances, covariances and means of DH and SSD populations so as to make it possible to conclude on the presence/absence of linkage. The suggested methodology is illustrated with an example concerning three quantitative traits of barley: length of the third internode, stem wall thickness, and 1000-grain weight.

The paper presents a proposition for detection of linkage of genes responsible for metrical traits. Taking into account the expected means for early generations (F1, F2, F3) and a population of homozygous lines (in this case doubled haploid lines, DH derived from a cross between two homozygous parents) as well as estimators of genetic parameters m, [d], [i], [h] and [l], the expected values for these parameters in the presence of linkage have been formulated. It was found that when there is no linkage, the expression F1 ? 6F2 + 8F3 ? 3DHmean is equal to zero. Thus, an experiment covering DH lines and F1, F2, F3 hybrids makes it possible to obtain, beside information of interest, also information on presence or absence of linkage.

Polymorphism of microsatellites S0083 and S0090 as well as of the second exon and second intron of the pGH (porcine growth hormone) gene was determined for 293 pedigrees obtained from crossing of 12 F1 (Zlotnicka Spotted x Polish Large White) boars with 64 F1 (Zlotnicka Spotted x Polish Large White) sows, experimental material arranged for a QTL mapping project. Microsatellites were genotyped by capillary electrophoresis of PCR products in an ABI PRISM 310 Genetic Analyser PERKIN-ELMER. The PCR-RFLP polymorphism of the GH gene was identified using HaeII and MspI restriction endonucleases. The following order of linked loci was established: GH-S0083-S0090. This is important information for the mapping of QTLs on chromosome 12.

Fire blight, caused by the gram-negative bacterium Erwinia amylovora (Burrill) Winslow et al., is a dangerous disease of pome fruits, including pear. A pear breeding program for fire blight resistance was initiated in 2003 at the Department of Pomology, Warsaw University of Life Sciences, Poland. Since several Asian species are considered to be potential sources of resistance to fire blight, the susceptible Pyrus communis'Doyenne du Comice' was crossed with the resistant P. ussuriensis. The F1 full-sib progeny composed of 155 seedlings was tested for susceptibility to fire blight by artificial shoot inoculation. A framework linkage map of both parents was constructed based on 48 AFLP and 32 SSR markers and covered a length of 595 cM and 680 cM in 'Doyenne du Comice' and P. ussuriensis, respectively. For the first time a putative QTL for fire blight resistance in P. ussuriensis linkage group 11 was identified. Another putative QTL in linkage group 4 of 'Doyenne du Comice' seems to indicate that sources of fire blight resistance can be identified also in the susceptible cultivars.
