Ocena stanu wiedzy młodych ludzi na temat HIV/AIDS

Introduction. Acquired Immunodeficiency Syndrome (AIDS) caused by human immunodeficiency virus (HIV) is one of the most serious infectious diseases on a global scale. Since the outbreak of the epidemic there was 78 million people infected with HIV, 35 million of whom have died. The AIDS control in Poland began in 1988. Aim. Assessment the knowledge level of young Polish people about AIDS. Materials and methods. The survey was conducted among 346 people utilizing the social media channels. They received 11 questions including the basic definitions, ways of infection and diagnosis of AIDS. Results. The results of our study confirm that people have insufficient knowledge of this serious disease. Less than 40% of all respondents could point the most common paths of infection and the time of first symptoms after exposure. Nearly 50% knew that AIDS and HIV are not equal terms. Conclusions. There is a need to raise awareness about HIV and AIDS. Young people should learn much more about mechanisms, symptoms and particularly prevention of this still untreatable disorder.

Wprowadzenie. Zespół nabytego niedoboru odporności, AIDS (ang. Acquired Immunodeficiency Syndrome), wywoływany przez ludzkiego wirusa niedoboru odporności (HIV, ang. human immunodeficiency virus) to jedna z najpoważniejszych chorób w skali świata. Szacuje się, że od wybuchu epidemii zakażeniu wirusem uległo 78 milionów ludzi, a około 35 milionów chorych zmarło. W Polsce epidemiologia i problematyka AIDS oraz HIV została podjęta w 1988 roku. Cel pracy. Ocena stanu wiedzy młodych ludzi na temat HIV i AIDS. Materiały i metody. Przeprowadzono ankietę internetową wśród 346 osób. Respondenci otrzymali 12 pytań na temat podstawowych pojęć, dróg zakażenia oraz diagnostyki AIDS. Wyniki. Wyniki ankiety potwierdzają, że wiedza młodych ludzi na temat HIV i AIDS jest niewystarczająca. Niecałe 40% ankietowanych potrafiło wskazać poprawnie główne drogi infekcji oraz prawidłowy przedział czasu pomiędzy ekspozycją a pojawieniem się pierwszych niespecyficznych objawów. Blisko połowa badanych miała świadomość, że pojęcia AIDS i HIV nie są tożsame. Wnioski. Istnieje potrzeba szerzenia wiedzy na temat HIV i AIDS, szczególnie wśród młodych ludzi. Tylko świadomość mechanizmu choroby, dróg zakażenia, a przede wszystkim możliwej profilaktyki, pozwoli na zminimalizowanie ryzyka zachorowania na chorobę, na którą obecnie wciąż nie wynaleziono remedium.

The aim of this study was to compare the efficiency of DNA extraction from water as well as from blood samples spiked with A. fumigatus spores, using selected commercial kits. Extraction of DNA according to manufacturer's protocols was preceded by blood cells lysis and disruption of fungal cells by enzymatic digestion or bead beating. The efficiency of DNA extraction was measured by PCR using Aspergillus-specific primers and SYBR Green I dye or TaqMan probes targeting 28S rRNA gene. All methods allowed the detection of Aspergillus at the lowest tested density of water suspensions of spores (101 cells/ml). The highest DNA yield was obtained using the ZR Fungal/Bacterial DNA kit, YeastStar Genomic DNA kit, and QIAamp DNA Mini kit with mechanical cell disruption. The ZR Fungal/Bacterial DNA and YeastStar kits showed the highest sensitivity in examination of blood samples spiked with Aspergillus (100 % for the detection of 102 spores and 75 % for 101 spores). Recently, the enzymatic method ceased to be recommended for examination of blood samples for Aspergillus, thus ZR Fungal/Bacterial DNA kit and QIAamp DNA Mini kit with mechanical cell disruption could be used for extraction of Aspergillus DNA from clinical samples.

Vitamin D3 (1,25(OH)2D3 (1,25-dihydroxyvitamin D3)) is a hormone playing a crucial role in numerous biological processes in the human body, including induction and control of cell proliferation and differentiation. Numerous data relate the vitamin D3 level with various types of cancer. It has been suggested that SNPs in the vitamin D3 receptor (VDR) gene might influence both the risk of cancer occurrence and cancer progression. The aim of this study was to search for genetic correlations between individual SNPs in the VDR gene and the risk of oral cavity carcinoma. Two SNPs were selected based on the literature and our previous results. Seventy-three patients with squamous cell carcinoma of the head and neck and one hundred control subjects were investigated. Two SNPs in the VDR gene were genotyped in minisequencing reactions followed by capillary electrophoresis. Hardy-Weinberg equilibrium (HWE), the χ2 test and logistic regression were used for statistical analysis. The SNP rs2238135 in the VDR gene displayed statistical differences in frequency between the tested groups (p=0,0007). Furthermore, the G/C genotype of the rs2238135 in the VDR gene was characterized by a 3.16 fold increased risk of oral cavity carcinoma. The obtained results provide evidence for a genetic association between rs2238135 in the VDR gene and the occurrence and risk of oral cavity cancer.
