It has been found that during the 14 days period of germinatlon and development of corn the RNA content in the whole root increased almost 4 times in comparison with the control root (2 days). These changes are statistically significant. The highest increase of the RNA content and the dry mass of the root has been observed during the first 6 days of development. The araount of rRNA and tRNA has changed in the same way. The rate of the increase of their content in comparison with the control root (2 days) was eąual and it suggest that in the tested period of development the rRNA and tRNA are synthetised with equal speed.

The fluorimetrio method with DABA x 2HC1 was applied in the work in localizlng the DNA in fractions of alkaline sucrose density gradient. The human lymphocytes used in the experiments were exclted towards the blastic transformation wlth phitohemaglutinine (PHA+), the unexoitated human lymphocytes (PHA-) and the commercise samcle of DNA izola ted frora the calf thymus (Sigma Chem. Co) were also usod. The obtalned data show that the results of the fluorimetric assay of the human lymphocytes DNA using the DABA x 2HCl in the fractions of the sucrose density gradient are in agreement with results obtained using the radioizotope method,' both for the unirradiated as well as for irradiated lymphocytes, and that this method might be used in the estimations of the molecular weight of the DNA using the centrifugation method in the sucrose density gradients without previous labelling of the cells.

In der vorliegenen Arbeit verwendete man die fluorymetrischen Methoden aus DABA x 2HCl zur Bezeichnung der DNS-Lokalisierung in den Fraktionen des alkalischen Gradients der Dichtigkeit der Sacharose. Man gebrauchte die menschlichen Lymphocyten, die zur blastlschen Transformation mit Hilfe von Fitohemaglutyninum (PHA+) angeregt wurden, diś nicht angeregten menschlichen Lymphozyten (PHA-) und das geziichtete Praparat der DNS des Kalbsthymus. (Sigma Chem. Co). Die gewonnen Angaben zeigten, dass die Ergebnisse der fluorymetrischen Kennzeichnung DNS der menschlichen Lymphocyten bei der Verwendung von DABA x 2HC1 in den Fraktionendes alkalischen Gradients der Dichtigkeit von Sacharose mit den Ergebnissen, die mit Hilfe der radioizotopischen Methode iibereinstimmen, sowohl im Fali der bestrahlten Lymphozyten ais auch der unbestrahlen. Diese Methode kann bei den Untersuchungen der Masse von Molekiilen der DNS mit Hilfe des Wirbelins in den Gradienten der Dichtigkeit der Sacharose verwendet werden,, ohne die Notwendichkeit der friiheren Kennzeichnung der Zellen.

During the 14 days period of development of the corn root the decrease of dry mass, ribosomal and Iow molecular RNA content was observed in the 10-mm root tip. The changes are statistically significant and they are of higher values during the first 4 days of germination. The continuous decrease of the dry mass content and RNA suggests that during the tested period of time the number of cells decreases, probably because of the reduction of the meristemic zone.
