Thyroid tumors are a very common disorder and can occur in six percent of human population. Despite this fact, there are still no effective and reliable techniques that would allow to pose a reliable differential diagnosis between malignant and benign thyroid tumor. The prevailing diagnostic techniques are: ultrasonography, scinthigraphy and fine needle aspiration cytology. In many cases they can not distinguish carcinomas from benign neoplasms. Such diagnosis is essential, because the patient with malignancy undergoes a very rigorous treatment that is unnecessary and inadvisable for patient with benign lesions. Therefore, the thyroid tumor markers are searched for. Generally tumor markers are substances, which presence or absence is related to malignancy. They can be used for population screening and for detection, diagnosis, staging, prognosis or follow up of malignant diseases. The thyroid tumor markers currently used have very restricted applications. The first one — calcitonine is produced only by one kind of cancer (medullary carcinoma) and the second — thyroglobulin is useful only in detection of recurrent follicular and papillary thyroid carcinoma. Therefore, there is a need to search for new tumor marker that could enable to differentiate benign lesions in thyroid from malignancies. This review article presents some information about thyroid neoplasms and methods of their diagnosis, highlighting current and possible usage of tumor markers.

Supported liquid membrane extraction of peptides.

The application of supported liquid membrane (SLM) extraction for the enrichment of short peptides is presented. The extraction efficiency is dependent on the pH of donor phase and salt concentration in acceptor phase. Moreover, the extraction efficiency is also influenced by the peptide amino-acid sequence and hydrophobicity.

Abstract: The objective of this work was to estimate glucose degradation products (GDPs) in solutions for peritoneal dialysis (PD) including glucose and sodium lactate based on the change of pH and absorption in UV. Spectrophotometric and pH-metric methods were used to measure glucose degradation and transformation of GDPs in laboratory-made solutions after heat sterilization and during storage. Mechanism of transformations of GDPs in the sterilized solutions for PD during storage and spectral characteristic of 5-HMF were studied. Common features for all the batches of the tested solutions for PD after heat sterilization were reduced pH, increase in the absorbance in the range 200-350 nm and the appearance of λmax at 274-283 nm. This indicated that 3,4-dideoxyglucosone-3-ene (3,4-DGE) and 5-hydroxymethylfurfural (5-HMF) had been formed during heat sterilization. Hypsochromic shift relative to the 5-HMF was explained by a spectral interference of levulinic acid having λmax at 266 nm. The change of pH after sterilization depended on the initial pH (before sterilization) and glucose concentration. During storage at room temperature hypsochromic or slight batochromic shift was observed. The absorbance at 228-230 nm diminished while that at λmax 270.5-280 nm was slightly reduced or even increased depending on the composition of a solution and time of storage. It was established that 5-HMF had two absorption maxima at the wavelengths of 228-229 and 283-284 nm in water medium and the absorbance of 5-HMF at 284 nm followed Beer’s law very well in the range of concentration of 1.91-9.85 mg/L.

An increase in the antibiotic resistance among Enterococcus faecium strains has been observed worldwide. Moreover, this bacteria has the ability to produce several virulence factors and to form biofilm that plays an important role in human infections. This study was designed to compare the antibiotic resistance and the prevalence of genes encoding surface protein (esp), aggregation substance (as), surface adhesin (efaA), collagen adhesin (ace), gelatinase (gelE), and hialuronidase (hyl) between biofilm-producing and non-producing E. faecium strains. Therefore, ninety E. faecium clinical isolates were tested for biofilm-forming ability, and then were assigned to two groups: biofilm-positive (BIO+, n =70) and biofilm-negative (BIO-, n = 20). Comparison of these groups showed that BIO+ isolates were resistant to β-lactams, whereas 10% of BIO- strains were susceptible to ampicillin (statistically significant difference, p = 0.007) and 5% to imipenem. Linezolid and tigecycline were the only antibiotics active against all tested isolates. Analysis of the virulence factors revealed that ace, efaA, and gelE genes occurred more frequently in BIO- strains (ace in 50% BIO+ vs. 75% BIO-; efaA 44.3% vs. 85%; gelE 2.9% vs. 15%, respectively), while hyl gene appeared more frequently in BIO+ isolates (87.1% BIO+ vs. 65% BIO-). These differences were significant (p < 0.05). We concluded that BIO+ strains were more resistant to antibiotics than BIO- strains, but interestingly, BIO- isolates were characterized by possession of higher virulence capabilities.

A novel diagnostic method based on recognition of qualitative and quantitative composition of healthy and tumor tissue samples by library of molecular receptors was presented. Molecular receptors were formed by self-organization of N-lipidated peptides attached in the regular fashion via aminophenylamino-1,3,5-triazine linker to the surface of cellulose plate. For samples testing, the library was cloned into multiple, identical copies and for each experiment the new clone was used. The binding process was monitored by staining the discs with Brilliant Black and quantitative color measurement was performed in 256 grade gray scale. Substantial differences in the composition of healthy and tumor samples were observed in most cases. The highly individual chemical fingerprints were found to be reliant on the cancer type. For malignant papillary thyroid cancer statistical analysis identified two receptors most useful for diagnostic purposes.
