The objective of the present work was to develop a simple and reliable protocol for chromosome doubling in haploid onion plants. Research on gynogenic plants obtained from different genotypes of onion was carried out over two years. In the first year, diploidization was examined in vivo with 2.5 mM colchicine for 24 h. In the next year, in vitro tests using 0.25 mM and 1.25 mM colchicine or 1.00 muM and 5.00 muM trifluralin were carried out. Both antimitotic agents were applied for 72 h at 14?C or 23?C. The obtained results showed that colchicine applied in vivo caused twice as high loss of then plant material than diploidization at in vitro conditions. Antimitotic agents added to culture media decreased the plant capability to regenerate. Colchicine was found to be less toxic than trifluralin. The use of colchicine allowed for producing more diploids than trifluralin.

An efficient method for genetic Agrobacterium tumefaciens - mediated transformation of five cultivars Gerbera hybrida was established. The youngest leaves and shoot tips from proliferating in vitro cultures were co-cultivated with disarmed strain LBA 4404/pBI121 carrying the chimaeric genes nptII and gus. The inoculated explants were repeatedly cultured on regeneration medium with kanamycin and Biotaxym. After 9-10 months from 0.04 to 0.25 independently transformed shoots from one inoculated leaf and from 0.06 to 0.65 from one inoculated shoot were obtained, depending on cultivar and additional treatment. Transformed shoots accounted for 5.0 to 50.0% of all regenerated shoots. Additional wounding` of leaves before inoculation by squeezing and scratching off increased transformation efficiency. Better results for recalcitrant to regeneration cultivars were obtained if squeezed shoot tips were inoculated. Regenerating shoots were selected on kanamycin, screened for gus expression with X-gluc and tested by PCR for nptII gene. All together 162 transgenic shoots were obtained in cvs Amber, Boy, Ferrari, Mariola and Tamara, but only 24 relatively stable lines were established.

Deformability and activity of the enzymes: acetylcholinesterase (AChE) and dehydrogenase glucose-6-phosphate (G-6-PD), were assayed for RBC enriched in immature reticulocytes. Reticulocytosis was evoked by administration of two different drugs: recombinant human erythropoietin (rHuEPO) and phenylhydrazine (PHZ) to two groups of Wistar rats. After treatment with the former compound, a group of animals exhibited 17,33 % reticulocytes in blood whereas a group of rats treated with the latter drug reached 57,66 % of these cells in blood. A marked decrease in RBC deformability was found in both groups of animals. AChE did not significantly change activity neither in PHZ-treated nor in rHuEPO-treated rats, whereas G-6-PD activity was significantly decreased in the PHZ-treated group.
